Di balik kesibukannya, Yayuk memiliki waktu-waktu khusus untuk menyalurkan kepeduliannya pada sesama lewat lembaga yang dinahkodainya. Ketua Lembaga Kemanusiaan (LK) ESQ Jawa Timur ini telah membuat program baik bidang pendidikan ataupun kesehatan. Salah satunya yaitu Home Care yaitu 22 program kunjungan ke rumah dan rumah sakit menemui para penderita kanker. Dalam setiap kunjungan, Yayuk membawa pasukannya yang terdiri dari relawan LK ESQ, karyawan Universitas Narotama termasuk mahasiswa. Tujuannya tidak lain agar mereka memiliki jiwa empati serta mensyukuri hidup.

Menutup wawancara Yayuk memberikan pesan untuk perempuan Indonesia, “Kita memiliki budaya saling menghormati. Saya ingin di Indonesia laki-laki dan perempuan saling menghormati. Jangan sampai ada paradigma laki-laki dikalahkan perempuan dan perempuan merasa dijajah oleh laki-laki. Itu sudah tidak zamannya, perempuan Indonesia berkaryalah. Berikan kontribusi nyata bagi dunia. Perempuan kelahiran Ponorogo itu mengingatkan bahwa orangtua jaman dulu meskipun minim pengetahuan kesehatannya, mereka ternyata mampu menyusui anaknya hingga dua tahun penuh. Mereka memberikan ASI eksklufif kepada bayinya selama 6 bulan.

Sebagai orang nomor satu di Kabupaten Probolinggo, Puput Tantriana Sari juga melarang semua instansi kesehatan di wilayah ini mulai rumah sakit umum daerah (RSUD) hingga Puskesmas menyediakan susu formula.

Bagaimana kiat membagi waktu antara karir dan keluarga? Menurut dia, terpilih menjadi bupati merupakan sebuah amanah yang harus dijalani dan dipertanggungjawabkan. Dia pun menjalani amanah tersebut dengan enjoy, belajar mengatur waktu antara jabatan (bupati) dan sebagai ibu rumah tangga. Kiat untuk menjaga keseimbangan tersebut dengan berusaha adil. Adil di sini bukan mengalokasikan waktu 50% untuk kantor dan 50% untuk rumah atau keluarga. Adil dalam pengertian kualitas, yakni kalau di kantor untuk urusan kerja pemerintahan. Sedangkan di rumah untuk urusan keluarga, melayani suami dan anak-anak.
